New compounds obtained by replacing benzene rings with pyridine tend to have higher biological activity or lower toxicity. Methyl 5-bromopyridine-2-carboxylate belongs to this type of compound, which is often used as an intermediate in pharmaceutical synthesis. |
| preparation method | add 60 mL of 65% sulfuric acid and 40 mmol of pyridine -2-carboxylate methyl ester into a 100 mL four-mouth bottle equipped with a mechanical stirrer, a constant pressure drop funnel and a thermometer, and slowly add NaBrO3 (6.0g,40 mmol) under stirring, the temperature is controlled and the reaction is cooled at 25~35 ℃ for 4h. The reaction mixture is placed in an ice bath and neutralized with a dilute alkali solution until alkaline. Extraction with dichloromethane (15 mL × 3), anhydrous MgSO4 drying, filtering, distilled to remove solvent, reduced pressure distillation to collect fractions to obtain methyl 5-bromopyridine-2-carboxylate. |
